Regeneration of chrysanthemum (Chrysanthemum morifolium) via somatic embryogenesis and screening of clones for agronomic traits

چکیده
Chrysanthemum morifolium propagation through stem cutting produces weak plants that show delayed anthesis above 20 °C and a reduced flower diameter. Therefore, in this study chrysanthemum plantlets were produced somatic embryogenic from calli to exploit the somaclonal variation for their improvement. Various explants variety Dante yellow cultured on Linsmaier Skoog (LS) medium with various concentrations of Kinetin (KT) 2,4-dichlorophenoxyacetic acid (2,4-D) combinations callus induction. Embryogenic proliferated regenerated by using different plant growth regulators. Regenerated acclimatized evaluated agronomic characteristics compared mother replicated field trial. Results revealed young leaf LS containing 9.02 µM 2,4-D 11.61 KT gave greater sizes as other treatments. A combination 0.44 6-benzylaminopurine (BAP) 5.37 1-naphthaleneacetic (NAA) yielded highest proliferation (0.27 g ± 0.03). significant number shootlets (25 0.8) was observed 0.45 2,4-D. During experiment, Clone S84 showed considerable improvement size found be promising clone commercialization. Somaclonal can used crop instead induced mutagenesis. Present substantial improved diameter via embryogenesis. Field screening increase flowers colour population. Both features are appealing consumers floriculture industry.

Identification of Chrysanthemum (Chrysanthemum morifolium) Self-Incompatibility
There has been a heated argument over self-incompatibilityof chrysanthemum (Chrysanthemum morifolium) among chrysanthemum breeders. In order to solve the argument, we investigated pistil receptivity, seed set, and compatible index of 24 chrysanthemum cultivars. It was found that the 24 cultivars averagely had 3.7-36.3 pollen grains germinating on stigmas at 24 hours after self-pollination throu...
متن کاملMicropropagation of Chrysanthemum (chrysanthemum Morifolium) Using Shoot Tip as Explant
Chrysanthemum has been cultivated for more than 2000 years and today, it is the world’s second most economically important floricultural crop following the rose (Teixeira da silva, 2003). Due to high popularity and demand for chrysanthemum it has become one of the first commercial targets for Micro propagation and thus tissue culture can be utilized for its large scale production. Regeneration ...
متن کاملEfficient In Vitro Regeneration of Chrysanthemum (Chrysanthemum morifolium Ramat.) Through Nodal Explant Culture
An efficient in vitro plant regeneration protocol was developed for Chrysanthemum morifolium, an ornamental plant. Nodal segments were used as explants in the present experiment. The explants were cultured onto Murashige & Skoog’s (MS) nutrient medium containing plant growth regulators (cytokinins or auxins) with various combinations and concentrations for the study. Maximum percentage (93.33%)...
متن کاملAntimutagenic activity of flavonoids from Chrysanthemum morifolium.
A methanol extract from the flower heads of Chrysanthemum morifolium showed a suppressive effect on umu gene expression of the SOS response in Salmonella typhimurium TA1535/pSK1002 against the mutagen 2-(2-furyl)-3-(5-nitro-2-furyl)acrylamide (furylfuramide). The methanol extract was re-extracted with hexane, chloroform, ethyl acetate, butanol, and water. The ethyl acetate fraction showed a sup...
متن کاملReference Gene Selection for RT-qPCR Analysis of Flower Development in Chrysanthemum morifolium and Chrysanthemum lavandulifolium
Quantitative real-time PCR (qPCR) is a popular and powerful tool used to understand the molecular mechanisms of flower development. However, the accuracy of this approach depends on the stability of reference genes. The capitulum of chrysanthemums is very special, which is consisting of ray florets and disc florets. There are obvious differences between the two types of florets in symmetry, gen...
